Flagella Experiment

To determine which flagellar component was most important, the following experiment was performed.

  • Cells are transposon tagged.

  • Cells are incubated with an attractant at the top of the flask.

  • Cells at the bottom are isolated and incubated in a flask with an attractant at the top.

  • Cells at the bottom are isolated and incubated in a flask with an attractant at the top.

  • Cells at the bottom are plated.

  • Cells from individual colonies are examined for morpholological differences from wild type cells.

  • Result: all mutants lacked radial spokes.

There is another experiment possible, although it is not as reliable, where you screen for small colonies after transposon tagging.
