Flagella Experiment

To determine which flagellar component was most important, the following experiment was performed.
Cells are transposon tagged.
Cells are incubated with an attractant at the top of the flask.
Cells at the bottom are isolated and incubated in a flask with an attractant at the top.
Cells at the bottom are isolated and incubated in a flask with an attractant at the top.
Cells at the bottom are plated.
Cells from individual colonies are examined for morpholological differences from wild type cells.
Result: all mutants lacked radial spokes.
There is another experiment possible, although it is not as reliable, where you screen for small colonies after transposon tagging.
